AFLP Analysis of Genetic Diversity of 30 Tree Peony (Paeonia suffruticosa Andr.) Cultivars

Abstract: 【Objective】Investigation of the tree peony's genetic diversity (Paeonia suffruticosa Andr.) is significant in utilizing its germplasm resources and cultivating new varieties. 【Method】The phylogentic relationships among 30 tree peony cultivars were analyzed using AFLP to study the genetic diversity with eight AFLP primer combinations (M+3 and P+3) . 【Result】Among the 1123 bands detected,965 bands were polymorphic and specific bands were found in all 30 tree peony cultivars. The high ratio of polymorphic bands (86%) observed in this study indicate abundant genetic diversity among tree peony cultivars. 【Conclusion】Our results suggest that the 8 pairs of prime combinations could be used to distinguish between all cultivars being tested. Results of cluster analysis showed that the classification based on AFLP markers of cultivars was consistent with their origin.
