海岛棉DNA导入陆地棉栽培品种获得变异种质的初步遗传分析

Genetic Analysis of Upland Cotton Germplasm Obtained from Introduced DNA from Island Cotton

摘要: 利用花粉管通道导入技术将海岛棉(Gossypium barbadense L.)品种海7124的基因组总DNA导入陆地棉(G. hirsutum L.)栽培品种石远321中,获得了纤维品质优良的新种质系,并且在导入后代中发现了1个形态性状发生变异的突变体。对突变体进一步的遗传分析表明,突变性状可能受1个纯合致死的显性基因控制,利用SSR对变异材料进行研究发现,外源DNA已经整合在陆地棉的基因组中,并推测某些DNA片段可能是通过同源重组的机制整合到陆地棉染色体上的。

Abstract: The genomic DNA of island cotton (Gossypium barbadense L. cv. Hai7124) was introduced into the upland cotton (G. hirsutum L. cv. Shiyuan321) by pollen tube pathway technique. New germplasm of high quality of fiber and a mutant with obviously change of morphology were obtained. Genetic analysis of the mutant indicated that the phenotype of the mutant was possibly controlled by a dominant allele that has homologous lethal effect. The result of SSR (simple sequence repeats) analysis showed that at least some exogenous DNA fragments were integrated into the genome of upland cotton Shiyuan321. It is deduced that some of the DNA fragments were integrated by homologous recombination mechanism.
