单抗介导牛海绵状脑病免疫组化检测方法的建立及其应用

摘要: 中国鲁西黄牛重组PrPc成熟蛋白免疫PrPc基因敲除小鼠,利用淋巴细胞杂交瘤技术,经免疫组织化学试验筛选出单抗4C11可特异识别牛海绵状脑病标准阳性牛脑样品上PrPSc核心片段。以单抗4C11为核心试剂,建立了单抗介导牛海绵状脑病免疫组织化学检测方法,并已颁布成为中华人民共和国国家标准GB/T19180-2003(牛海绵状脑病诊断技术)。已应用于中国牛海绵状脑病的持续监测,至2002年底共检测了全国各地3344份牛脑样品,结果全为阴性,且每批次所设的阳性对照样品检测结果均呈阳性,与Roche公司单抗6H4检测结果的符合率为100%。

Abstract: One hybridoma cell line 4C11 secreting specific antibodies to core part of PrPSc in Bovine Spongiform Encephalopathy (BSE) were established from several fusions between mouse myeloma Sp2/0 and spleen cells from mice immunized with recombinant Chinese Luxi yellow cattle PrPc mature protein. An immunohistochemistry assay based on monoclonal antibody 4C11 for the detection of BSE was established and has been applied for the long-term surveillance of BSE in China. By the end of 2002, 3 344 samples from the whole country had been detected and all had negetive results.
