Expression of Ghrelin and GHS-R1a mRNA in the Bovine Oocytes During in Vitro Maturation

摘要: 【目的】探讨Ghrelin和GHS-R1a mRNA在牛卵母细胞体外成熟过程中的表达及二者表达与FSH之间的关系。【方法】 利用实时定量RT-PCR技术检测体外成熟进程中牛卵母细胞Ghrelin和GHS-R1a mRNA水平表达变化,观察不同浓度FSH对Ghrelin和GHS-R1a mRNA表达的作用以及FSH与FSH受体抑制剂的组合对Ghrelin和GHS-R1a基因表达的影响。【结果】 实时定量RT-PCR结果揭示牛卵母细胞Ghrelin和GHS-R1a mRNA的相对表达量随着成熟过程的延长而呈现一定变化规律,即Ghrelin和GHS-R1a mRNA在卵母细胞体外成熟过程中从8 h开始显著下降并持续此低表达水平到24 h;不同浓度FSH 显著抑制卵母细胞Ghrelin及GHS-R1a mRNA的表达,而FSH 受体抑制剂明显减弱FSH的抑制作用而促进二者表达。【结论】Ghrelin和GHS-R1a mRNA在卵母细胞体外成熟过程中的表达可能随着培养液中FSH升高而降低。

Abstract: 【Objective】 The aim of this study was to investigate the possibility of Ghrelin and GHS-R1a mRNA expression in bovine oocytes produced in vitro and the relationship between FSH and the expression of Ghrelin and GHS-R1a mRNA. 【Method】Relative real time RT-PCR technology was applied to detect the pattern of Ghrelin and GHS-R1a mRNA expression in oocytes, and the effect of different concentrations FSH and the combination of FSH and FSH binding inhibitor on the expression of Ghrelin and GHS-R1a mRNA were observed. 【Result】 Relative real time RT-PCR experiments confirmed that the Ghrelin and GHS-R1a mRNA levels in oocyte varied depending on the developmental stage, with the significant decrease of Ghrelin and GHS-R1a mRNA expression at 8 h in oocyte maturation process. And addition of FSH inhibited the expression of Ghrelin and GHS-R1a mRNA within oocytes, and this inhibition effect could be attenuated by FSH binding inhibitor. 【Conclusion】 The expression of Ghrelin and GHS-R1a mRNA could be declined with the rising of FSH concentration during in vitro maturation of bovine oocytes.
