Identification of a multidrug-resistant porcine Proteus vulgaris harbouring a plasmid-borne cfr gene

 Highlights 
A novel conjugative plasmid pHJ90-cfr carrying the multiresistance gene cfr was characterized in Proteus vulgaris.
A new IS5-family member, ISPmi4, was identified for the first time.
Both plasmids and ICEs were vital mobile genetic elements for horizontal transmission of cfr gene in Proteus species.

摘要  

噁唑烷酮类抗生素作为目前治疗多重耐药革兰氏阳性菌感染最有效的抗生素之一,其耐药基因cfr的流行和传播备受关注,然而,目前cfr基因在普通变形杆菌中的流行情况尚不十分清楚。本文从黑龙江省某猪场分离出一株多重耐药普通变形杆菌(Proteus vulgaris P. vulgaris)HJ90,通过肉汤稀释法测定了该分离株的药物敏感性,其次利用IlluminaNanopore测序技术测定了该分离株的全基因组序列,通过生物信息学技术对全基因组序列进行比对分析,随后利用接合转移试验检测质粒pHJ90-cfr的可转移性,最后利用PCR方法检测插入序列IS26介导的易位单元(Translocatable UnitTU),以进一步探究其在耐药性传播中的作用。研究结果显示,分离株HJ90表现出多重耐药表型。全基因组测序结果表明,成功获得了该分离株的完整染色体序列和两个质粒序列。耐药基因检测分析发现,该分离株的一个质粒pHJ90-cfr只携带cfr基因,而染色体及另一个质粒pHJ90-1则携带多个其它类型抗生素的耐药基因。接合转移试验结果表明质粒pHJ90-cfr可以成功转入受体菌Escherichia coli C600,且接合子表现出耐药表型。耐药基因环境分析显示cfr所在的耐药区域侧翼为两个同向的IS26插入序列,PCR检测结果显示携带cfr的耐药区域能够利用TU的方式进行水平转移,进而导致cfr的广泛传播。此外,本研究还发现并命名了一种新的IS5家族插入序列ISPmi4。总的来说,本研究在P. vulgaris中鉴定出一种新的可水平转移的耐药质粒pHJ90-cfr,该质粒携带多重耐药基因cfr。此外,还发现了一种新的IS5家族成员ISPmi4。本研究强调了cfr基因在革兰氏阴性菌中的流行情况需要进一步监测。
