番茄抗叶霉病基因Cf6的RAPD及SCAR标记

摘要: 【目的】寻找与番茄抗叶霉病基因Cf6连锁的分子标记。【方法】 以番茄抗叶霉病和感叶霉病亲本组合(03036×03748)的F2分离群体为研究材料,采用BSA法和RAPD技术筛选与抗病基因连锁的分子标记。【结果】经F2单株分析,在后代群体中扩增出了两条长约500 bp的特异片段,该特异标记与叶霉病抗性基因Cf6紧密连锁,遗传距离为8.7 cm。测序结果显示,目标片段的大小分别为619 bp和559 bp,并将该标记转换为SCAR标记。【结论】SCAR标记,可用于番茄叶霉病抗性分子标记辅助育种。

Abstract: Abstract: 【Objective】The study screened the molecular marker linked to the tomato Fulvia fulva resistant gene.【Method】With a F2 population between a resistant and a susceptible parent (03036×03748), the linked marker was screened by BSA method and RAPD technology.【Result】Two special bands were found at 500bp or so using F2 plants. The marker was tightly linked to the tomato Fulvia fulva resistance gene, and the genetic distance was 8.7cM. Sequencing of the fragment indicated that the length was 619bp and 559bp respectively.【Conclusion】The study provided a molecular method to identify tomato Fulvia fulva resistance , and can be used in molecular marker assisted selection.
