Adjuvanting a subunit novel variant IBDV vaccine to induce protective immunity

 Highlights 

Evaluated the safety and efficacy of subunit vaccine prepared with different adjuvants in chicks.
The vaccine prepared using the evaluated adjuvant has enhanced its protective effect against IBDV infection.
Gel P vaccine is the most convenient to prepare, and ISA 78 VG has better stability than other adjuvants.

摘要  

鸡传染性法氏囊病(Infectious Bursal Disease, IBD)是一种由鸡传染性法氏囊病毒(Infectious Bursal Disease Virus, IBDV)引起的急性、高度传染性疾病,主要危害雏鸡。新型变异株的出现,加剧该疾病对家禽业可造成巨大的经济损失,但现无针对新型变异株的商品化疫苗。目前,免疫预防是控制该疾病的主要手段。本研究比较了用白油佐剂、MontanideISA 78 VGMontanideGel P和氢氧化铝佐剂以及无佐剂制备的新型变异株IBDV VP2亚单位疫苗的安全性和有效性,通过组织解剖、病理切片观察、抗体水平检测、组织病毒载量检测等试验,评估对各种佐剂制备的疫苗的保护作用。结果表明,本研究制备的IBDV VP2亚单位疫苗能够诱导特异性抗体的产生,抑制法氏囊萎缩并抵抗病毒攻击。在抗原量(IBDV)相同的条件下,MontanideISA 78 VGMontanideGel P和氢氧化铝佐剂制备的疫苗保护明显优于其他佐剂和无佐剂组,其中MontanideISA 78 VG和氢氧化铝佐剂制备的疫苗效果最好,MontanideISA 78 VG佐剂疫苗最稳定,MontanideGel P佐剂疫苗最容易制备。综上所述,在安全性优先的情况下,使用MontanideTM ISA 78 VGMontanideTM-Gel P和氢氧化铝佐剂可以提高雏鸡疫苗免疫的安全性和有效性,为后续IBDV的防治提供实验参考和理论依据
