Infectious disease-resistant pigs: Will they fly?

摘要  

该评论文章认为,Xu等研究论文的重要意义在于,使用基因编辑技术在大白猪胎儿成纤维细胞中同时对猪CD163pAPN基因进行了编辑,精准删除了介导PRRSV和TGEV入侵的猪受体基因,使病毒不能进入猪体内,从而有效预防这些疾病。通过扩繁,获得了大量基因型一致的双基因编辑大白猪。猪活体攻毒实验表明,双基因编辑猪可以完全抵抗PRRSV和TGEV的感染,而未经编辑的野生型对照猪中均能建立感染并导致其患病。此外,研发人员还发现双基因编辑猪能显著性抑制PDCoV的感染,首次提供了pAPN作为PDCoV受体之一的活体实验证据。最后,通过长期的饲养观察及繁殖实验和国家种猪测定中心的屠宰实验等表明,双基因编辑猪的繁殖及生产性能正常。评论文章认为,该文通过基因组编辑技术对家畜成功进行多基因修饰并获得复杂性状的遗传改良,展示了新兴的基因组编辑技术在动物育种中的巨大应用价值,并为解决重大传染性疫病问题提供新的途径。同时,该双基因编辑猪也为抗重大疫病病猪新品种培育提供了重要的育种材料。
