1. High potassium to magnesium ratio affected the growth and magnesium uptake of three tomato (Solanum lycopersicum L.) cultivars

Potassium (K) and magnesium (Mg) levels and their balances are two factors affecting the growth of plant.  However, the responses of different crop cultivars to K/Mg ratios are less clear.  This study was aimed at assessing the different responses of tomato (Solanum Lycopersicum L.) cultivars to the different K/Mg supply ratios.  Three tomato cultivars (Zhongza 9 (ZZ), Gailiangmaofen (MF), and Jinpengchaoguan (JP)) were grown in pots with three different K+/Mg2+ ratios (4:0, 4:1 and 8:1, represented by K/Mg4:0, K/Mg4:1, and K/Mg8:1, respectively).  Compared with K/Mg4:1 treatment, the leaf chlorophyll content, net photosynthetic rate, and total biomass of tomato seedlings under K/Mg4:0 treatments were decreased by 69.7, 89.1, and 53.1%, respectively.  The Mg deficiency symptoms were observed when the Mg content in shoot became lower than 4 mg g–1 DW.  Compared with K/Mg4:1 treatment, total biomass of tomato seedlings of K/Mg8:1 treatment was decreased by 21.6%; the shoot and root Mg contents were decreased by 10.4 and 21.8%, respectively; and Mg uptake of tomato was reduced by 34.1%.  There were significant differences in biomass and Mg uptake for the three cultivars between the different K+/Mg2+ treatments.  The Mg uptake of the three different cultivars ranked as ZZ>JP>MF under Mg deficiency and high K condition.  In conclusion, the growth and Mg uptake and allocation of tomato were influenced significantly by imbalance K and Mg supply. JP and ZZ were the cultivars with the highest efficiency in Mg uptake.

2. Transgenic Expression of a Functional Fragment of Harpin Protein Hpa1 in Wheat Represses English Grain Aphid Infestation

The harpin protein Hpa1 produced by the rice bacterial blight pathogen promotes plant growth and induces plant resistance to pathogens and insect pests. The region of 10-42 residues (Hpa110-42) in the Hpa1 sequence is critical as the isolated Hpa110-42 fragment is 1.3-7.5-fold more effective than the full length in inducing plant growth and resistance. Here we report that transgenic expression of Hpa110-42 in wheat induces resistance to English grain aphid, a dominant species of wheat aphids. Hpa110-42-induced resistance is effective to inhibit the aphid behavior in plant preference at the initial colonization stage and repress aphid performances in the reproduction, nymph growth, and instar development on transgenic plants. The resistance characters are correlated with enhanced expression of defense-regulatory genes (EIN2, PP2-A, and GSL10) and consistent with induced expression of defense response genes (Hel, PDF1.2, PR-1b, and PR-2b). As a result, aphid infestations are alleviated in transgenic plants. The level of Hpa110-42-induced resistance in regard to repression of aphid infestations is equivalent to the effect of chemical control provided by an insecticide. These results suggested that the defensive role of Hpa110-42 can be integrated into breeding germplasm of the agriculturally significant crop with a great potential of the agricultural application.

3. The Influence of Bt-Transgenic Maize Pollen on the Bacterial Diversity in the Midgut of Chinese Honeybees, Apis cerana cerana

Using culture-independent technique polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) and conventional culture techniques, ecological risk of transgenic maize pollen on gut bacteria of the Chinese honeybee, Apis cerana cerana, was assessed. Honeybees were fed with Bt-transgenic maize pollen, non-transgenic near isoline pollen, linear cry1Ah gene (800 ng mL-1) and supercoiled plasmid DNA (800 ng mL-1) under laboratory conditions. The DGGE profile showed that the number of DGGE bands varied from 10.7 to 14.7 per sample, and the Shannon’s index ranged from 0.85 to 1.00. The similarity calculated by PAST was mostly above 92%, indicating no obvious changes among treatments or within replicates. 14 bacterial strains affiliated with Alphaproteobacteria, Gammaproteobacteria, Firmicutes, and Actinobacteria were isolated and characterized on media under aerobic and anaerobic conditions. These results demonstrated that transgenic cry1Ah maize pollen did not induce significant changes of the honeybee gut bacterial community composition under laboratory conditions.
